Sirtuins are a family of seven NAD+-dependent enzymes (SIRT1-SIRT7) that regulate cellular stress response, metabolism, and DNA repair. Each sits in a different compartment - nucleus, mitochondria, cytosol - and tunes gene expression in response to energy state. Rodent data ties sirtuin activity to extended lifespan, especially SIRT1 and SIRT6; human translation is promising but not conclusive. Raising NAD+ with NR or NMN is one of the cleanest ways to support sirtuin function, which is the premise behind most of the longevity supplement market.
